Paracetamol inhibits prostaglandin synthesis in intact cells in vitro when the levels of the substrate arachidonic acid are low, then prostaglandins are primarily synthesized by COX-2 in cells that contain both COX-1 and COX-2.
Analgesic effect of paracetamol is central due to the activation of descending serotonergic pathways, but its primary site of action is the inhibition of Prostaglandin synthesis.
